The Sharingan is not automatically activated in the user by birth. It is only first unlocked under some stressful or emotional condition after which the Sharingan may be used freely. When the Sharingan is first activated by a user, it has a different number of tomoe around the central pupil, varying between users.

The Mangekyō Sharingan gives the user access to several powerful techniques, including the powerful fire technique Amaterasu, the use of Susanoo and the almost unbreakable genjutsu Tsukuyomi. The Mangekyō Sharingan however erodes the eyesight of its users, eventually causing blindness, which can be prevented by obtaining another pair of Mangekyō Sharingan from a sibling.

Background:Following the Nine-Tails' attack on Konoha orchestrated by Tobi, Konoha's leadership began to suspect that an Uchiha was behind the attack because of the Sharingan's ability to control the Nine-Tailed Demon Fox. In order to keep an eye on the Uchiha, Tobi claimed they were forced to live in a corner of the village and kept under surveillance, creating dissent within the clan. When members of the clan began planning a coup d'état, they tasked Itachi Uchiha with spying on Konoha from within the ANBU. However, Itachi's pacifistic nature led him to become a double agent, instead providing Konoha with information on the Uchiha. He knew that if the suppressed, but powerful, Uchiha were to attack Konoha, the resulting civil war would weaken the village in all aspects. This sign of weakness would entice other countries to take advantage and invade Konoha, with the conflict eventually escalating into another Shinobi World War. As the Uchiha came closer to a village-wide revolt, the Third Hokage attempted to open negotiations with them. Unfortunately, time ran out and Hiruzen's advisers ordered Itachi to slaughter his entire clan.
Prior to the massacre, Itachi confronted Tobi, who had infiltrated the village. Madara, having decades earlier been shunned by the clan, still held resentment for both the Uchiha and Konoha. Itachi offered Tobi the chance to help him take his revenge against the clan, in exchange for promising not to attack Konoha, a deal which Tobi accepted. It was also during this time that Tobi met with Danzō Shimura; the circumstances of this meeting are unknown as they did not meet again until after the Kage Summit.
The Uchiha clan, however, was not completely blind towards Itachi's double-agent status. They began to suspect him, several months after Itachi had joined the ANBU, due to his change in behaviour. They ordered Shisui Uchiha, Itachi's closest friend, to keep an eye on him, however Shisui later turned up dead, seemingly committing suicide by drowning. The clan suspected that Itachi may have had something to do with this death, including the possibility of having forged the suicide note. Because of this, along with Itachi acting as though entering the ANBU was nothing special, the clan began to become antagonistic towards Itachi. Indeed Itachi had murdered Shisui in order to obtain his Mangekyō Sharingan and had forged the suicide note. However, he denied involvement at the time, and due to his mission of stopping the revolution being secret, he instead acted disrespectful towards his clansmen, as though he was beginning to lose faith with the clan's incessant need for their rules and regulations. It was later revealed that Itachi did not in fact murder Shisui, but instead aided him in his orchestrating his death, as Shisui was also a firm believer in maintaining the peace and prosperity of the village. Even Fugaku Uchiha, the clan head and Itachi's father, had lost hope in Itachi altogether, and the two had a falling out.
That was when Itachi and a unknown accelent had killed everyone of the uchiha's in the village other then sasuke and a few others.

Background Information:not much is known about alexander's past until after he was old anoth to kill at the age of 10 years old a academy student he had killed his first enemy ninja when the ninja had tried to kill his aunt and uncle right in front of him which is what unlocked his sharingan after he past the exams he was made a Genin and went on alot of dangerous missions even though their missions were supposed to be D level missions his was B level.
when he was 12 the held the chunnin exams which is where with his mind and strength he was made a chunnin.
From that time he trained and trained until he had was 16 he now had three tomoe per eye was a Jounin because of his incredible skills and the deady missions he always went on.
at the age of 18 he went to war with two friends of his a girl he loved and a guy which he knew since he was little. One night him and the girl where talking and his other friend was stairing at them he was acting funny but alexander paid no attention to it his mind was on the war which he didnt know when it would end but it would be soon him being the jounin he told them to sleep he then heard rustlings above him and saw his friend over him with a kunai with out thinking he activated his sharingan and moved quickly he stabbed his friend in the chest with a kunai killing him.
When he found out his friend had been under genjutsu he was saddened then anger went through him as he wanted revenge which activated his Mangekyō Sharingan.
At the age of 19 the war was finally over but alexander still wasnt happy him and the girl had survived luckly not that wouldnt whem they got back thanks to their good deeds they were honored him and the girl then got married but it wasn't a a year later her and the hokage where killed by a unknown assassin at the time until alexander returned at the time from a mission he found his wife and the hokage dead out of anger he tracked the killer down using his Space–Time Migration technique looking around for anyone that didnt belong when he found the assassin he killed him finding out he was from the village hidden in the mist he raised the alarm and told them that the hokage and his wife were dead he had the body on his shoulder he threw it down telling them that it was a assassin from the village of the mist.
The next morning a meeting was held for the choice of a new hokage they had two people in mind him and another Jounin but at the end they chose him because he would be a stronger leader.
